IN VITRO MORPHOGENESIS THROUGH GA3-TREATED SPATHE OF ANTHURIUM CARNAVAL LIND.

The spathe of Anthurium carnaval have been transformed in morphology and function like normal leaves when treated with GA3. Callus induction and plantlet regeneration of Anthurium carnaval via somatic embryogenesis derived from morphologically changed spathe were investigated. The results showed the change morphology with leaf-like of spathe was obtained by treating with 1 mg l-1 GA3. High frequency compact callus induction from green spathe explants (81.7%) was obtained on half-strength MS medium supplemented with 0.045 mg l-1 TDZ, 0.5 mg l-1 BA, under the dark condition after 60 days of culture. On half-strength MS medium supplemented with 1.0 mg l-1 2,4-D, 0.5 mg l-1 KIN, and 1000 mg l-1 myo-inositol, both the rate of embryo induction (83.3%) and number of embryos formed (18.7 embryos) were highest. The highest number of embryo-derived plants (15.3 plants) was observed after 90 days of culture. Genetic fidelity assessment showed complete monomorphisms the visible bands of the stock plant and 10 identical clones. This output confirmed the genetic homogeneity among the analyzed plantlets. This is the first report of plantlets regeneration from somatic embryogenesis derived from spathe change morphology.
