MICROPROPAGATION OF PAEONIA OSTII ‘FENG DAN’ VIA IN VITRO MATURE EMBRYO CULTURE

Paeonia ostii ‘Feng Dan’ is ornamental woody plant, but its breeding and commercial production is limited due to inefficient traditional propagation methods. The result of the experiments aimed at its propagation showed that modified Murashige and Skoog (MS) medium supplemented with 2.6 µM N6-benzyladenine (BA) + 2.9 µM Gibberellin acid (GA3) and modified woody plant medium (WPM) supplemented with 1.3 µM BA + 0.6 µM GA3 + 0.5 µM 6-Furfurilaminopurine (kinetin, KT) were the optimal media for embryo germination and shoot multiplication, respectively, with 8.1 shoots. Adding of 7.4 µM Indole-3-butyric acid (IBA) and 2.4 × 10-1 mM phloroglucinol in root induction medium notably enhanced the rooting rate (41.5%). Phloroglucinol was superior to coumarin and chlorogenic acid in promoting the height and survival rate of rooted shoots, with 2.4 × 10-1 mM phloroglucinol as the most effective (3.5 cm, 33.9%).
