EFFECTS OF MEDIUM PH AND CARBOHYDRATE SOURCE ON THE IN VITRO PROPAGATION OF THE ENDANGERED METALLOPHYTE DIANTHUS PINIFOLIUS SIBTH. ET SM.

The aim of this study was to evaluate the effect of medium pH and different concentrations of sucrose, fructose, and glucose on the regeneration and development of D. pinifolius axillary shoots on full- and half-strength Murashige and Skoog (MS) medium using different explant types. The results show that suitable pH for D. pinifolius growth in vitro is 5.8. Single nodes should be avoided in the multiplication phase, and terminal buds and shoots should be used as explants. They can be cultured on half-strength MS medium with pH 6.3 and 10 g l-1 sucrose, on MS medium with pH 5.8 containing 30 g l-1 sucrose, and on MS medium with 30 g l-1 glucose and pH 6.3. The rooting percentage was high on MS medium containing 0.5 mg l-1 NAA, 95% of single nodes and shoots and 96.7% of terminal buds formed roots. The highest acclimatization percentage of obtained microplants (95.5%) was achieved in a 4 : 1 mixture of peat and sand.
