Abstract
In our experiments on enhancement of in vitro regeneration potential of rose we have found that the addition of 2.5 mg l-1, TIBA, to the regeneration medium containing BAP intensified regeneration of adventitious shoots on leaves of rose cultivars White Gem, Mistral, and Charming and prolonged an efficient regeneration for the third and fourth subcultures. TIBA significantly enhanced the number of responding leaves and number of adventitious shoots. Effect of TIBA was counteracted by the addition of 0.01 mg l-1 NAA to the regeneration medium. Histological analyses showed that regeneration medium containing BAP and TIBA stimulated continuous cell division and formation of meristems and buds whereas on the medium without TIBA less of initiated meristems and differentiating buds but xylem clusters were observed.
