GLADIOLUS MICROPROPAGATION IN TEMPORARY IMMERSION SYSTEM

Abstract
Genus Gladiolus includes important ornamental species, and new hybrids have been recently selected with the aim to increase the production in winter and late spring in countries bordering the Mediterranean sea. In vitro culture was taken into consideration in order to obtain virus-free mother plants and set up a suitable propagation protocol. Meristem tips of 20 new hybrids were multiplied in MS medium containing 2iP (2.98 μM). After 3 months of shoot proliferation the explants were enlarged onto a medium containing IBA (4.90 μM). Cormels have been then transferred in vessels for Temporary Immersion Culture. Three minutes of flooding every three hours were set up and the vessels were cultured at 23ºC at a 16 h photoperiod at 35 μE m-2 s-1. Twelve weeks later the productivity and several quality parameters were evaluated. The cormels produced in TIS were very easily collected and then dried in laminar flow cabinet for 24 h and were stored at 5ºC for 6 months. After transplanting to soil an emergence of 100% was detected.
