IN VITRO BULBLET REGENERATION OF ORNITHOGALUM OLIGOPHYLLUM E. D. CLARKE USING TWIN SCALE BULB EXPLANTS

Abstract
In vitro bulblet regeneration of Ornithogalum oligophyllum E. D. Clarke.; a potential ornamental plant of Turkey was achieved using twin scales bulb explants on MS medium supplemented with combinations of 1-2 mg l-1 BAP and 0.5-1.0 mg l-1 IBA or on MS medium without plant growth regulators. The primary adventitious bulbs regenerated from twin scales were sub cultured on MS medium containing 1-2 mg l-1 BAP and 0.5-1.0 mg l-1 IBA. The sub cultured primary adventitious bulblets increased in size and regenerated 0 to 4.42 secondary bulblets per primary bulblet with roots of variable length. The highest adventitious bulblet regeneration was achieved on MS medium containing 2 mg l-1 BAP and 0.5 mg l-1 IBA. All plants were acclimatized in the environmental chamber and were transferred to the greenhouse for flowering. Regenerated bulblets developed into morphologically normal plants with survival rate of 85%. The results showed that the method could be successfully applied for the production of bulbs for field cultivation. The described procedure could also be used as an alternative for out of season and commercial rapid multiplication of the plant.
