IN VITRO REGENERATION AND PLANTLET FORMATION FROM ADVENTITIOUS ROOTS OF REHMANNIA GLUTINOSA LIBOSCHITS

Abstract
A novel method of direct shoot bud regeneration from adventitious roots in Rehmannia glutinosa was developed. The influence of different cytokinins on shoot initiation was examined. Among the different cytokinins tested in the present experiment BA was found to be the best inducer of organogenesis. The highest percentage of shoot formation (66.7 %) and the highest number of shoots per explant (7.0) were observed on MS media supplemented with 2 mg l-1 BA. However, shoot initiation and development were observed on growth regulator free media (66.7 % shoot regeneration with 6.3 shoots/explant). Rooting of individual shoots was induced after transfer onto half strength MS medium without growth regulators. Plantlets after acclimatization were successfully transplanted in the field and no phenotypic variations were observed among them. This technique has potential applicability for shoot biomass production from adventitious roots of Rehmannia.
