IN VITRO PRODUCTION OF PATHOGEN-FREE PLANT MATERIAL IN PRUNUS DOMESTICA L.

Abstract
An effective regeneration/multiplication system in selected cultivars of Prunus domestica was developed. The best culture medium for regeneration of meristems isolated from mature trees was 1/WPM supplemented with GA3 (0.1 mg l-1), BAP (1 mg l-1) and 2,4-D (0.002 mg l-1) inducing vital shoots with vigorous growth. The highest multiplication rate was achieved on 3/MS multiplication medium supplemented with GA3 (0.1 mg l-1), BAP (1 mg l-1) and IBA (0.1 mg l-1). In vitro propagation of Prunus domestica in combination with chemotherapy enables elimination of pathogens from plant tissue and production of pathogen-free plants.
