A protocol for adventitious shoot regeneration of Haworthia retusa has been established. Leaf explants were cultured on Murashige and Skoog (MS) medium supplemented with different concentrations of thidiazuron (TDZ) alone or in combination with 2,4-dichlorophenoxyacetic acid (2,4-D), indole-3-acetic acid (IAA), and α-naphthaleneacetic acid (NAA) in order to promote adventitious shoot regeneration. Of the varying concentrations of TDZ studied, the frequency of shoot induction (89.8%) with a mean of 12.7 shoots was observed to develop directly from leaf explants on MS medium amended with 4 µM TDZ. The inclusion of auxin to TDZ-containing medium significantly increased the shoot induction frequency as well as shoot numbers. TDZ + 2,4-D, TDZ + IAA, and TDZ+NAA combinations produced white fragile, greenish soft, and pale green nodular calluses, respectively. The best shoot induction (100%) and maximum shoot numbers (38.7) were obtained on explants cultured on a medium 4 µM TDZ and 2 µM NAA combinations. Rooting of shoots was best achieved on MS medium amended with 0.1% (w/v) AC and 2 µM indole-3-butyric acid.

Key words: auxin, callus formation, leaf explants, shoot regeneration, thidiazuron
