EFFICIENT IMMATURE EMBRYO GERMINATION IN VITRO
OF HICKORY (CARYA CATHAYENSIS SARG.)

Abstract
Hickory (Carya cathayensis Sarg.) is an ornamental or a shade tree for landscape. Due to its poor germination, conventional propagation through seeds and graft is not sufficient to satisfy the progressive demand. The purpose of this research was to standardize the media and culture conditions for organogenesis via immature embryo culture of hickory (Carya cathayensis Sarg.). The immature embryos of hickory were cultured on Murashige and Skoog medium. Significant variations were observed according to embryo age and plant growth regulators (PGRs). For embryo age, a high frequency (77.5%) of plantlet was obtained from the immature embryos sampled after 14 weeks post-pollination. When BA, picloram and GA3 were respectively added, significant difference was found in germination percentage, root length and seedling length. The highest percentage of germination (65.5%) was obtained on the medium supplemented with 2.0 mg l-1 BA. When the concentration of picloram was the highest (0.1 mg l-1), the root length was the highest and reached 2.6 cm. The seedling length was also the highest when GA3 was medium (0.5 mg l-1). Percentage of immature embryo germination was 90.3% when BA (2.0 mg l-1) and picloram (0.01 mg l-1) were applied simultaneously. Percentage of germination was only 71.9% when BA (2.0 mg l-1) and GA3 (1.0 mg l-1) were applied simultaneously.
