IN VITRO ADVENTITIOUS SHOOT FORMATION FROM PETIOLE EXPLANTS
OF SWAINSONA SALSULA TAUBERT

Abstract
An effective protocol was developed for adventitious shoot formation from petiole explants of Swainsona salsula Taubert. Our results showed that MS medium with 1.5 mg l-1 BA and 15 g l-1 sucrose was optimal for the induction of adventitious shoots (85%), while the highest shoot numbers per responding explant (an average of 4.2) were obtained on MS medium containing 2.0 mg l-1 BA and 30 g l-1 sucrose, but with high hyperhydricity. Three experiments for preventing hyperhydricity were studied. The results showed that sealing the conical flasks (150 ml) with cotton plugs could significantly decrease the hyperhydricity rate from 36% to 9%, and meanwhile, increase the shoot induction rate from 53% to 77%. When individual elongated shoots were inoculated on half strength MS medium supplemented with 2.0 mg l-1 IBA, 74% successfully rooted.
